黄芪注射液对缺血后脑组织神经干细胞增殖和分化的影响 被引量:25

摘要 目的探讨黄芪注射液对缺血后脑组织神经干细胞增殖和分化的影响。方法 45只Wistar大鼠随机分为假手术组、对照组和黄芪组;通过夹闭大脑中动脉建立脑缺血模型。黄芪组于建模前12 h及建模后1~10 d每天腹腔注射黄芪注射液(10 ml/kg);假手术组和对照组腹腔注射等量生理盐水。建模3 d、7 d、10 d应用免疫组化和图像分析技术检测脑组织巢蛋白(Nestin)及其与星形胶质纤维酸性蛋白(GFAP)或微管相关蛋白2(MAP2)共同表达的阳性细胞数。结果 (1)与假手术组比较,对照组和黄芪组Nestin阳性细胞数显著增高(均P<0.05);黄芪组各时间点Nestin阳性细胞数显著高于对照组(均P<0.05)。(2)黄芪组各时间点Nestin-GFAP及Nestin-MAP2阳性细胞数显著多于对照组(均P<0.05)。(3)对照组和黄芪组各时间点Nestin-GFAP阳性细胞数明显多于Nestin-MAP2阳性细胞数(均P<0.05)。结论黄芪注射液能促进脑组织Nestin表达,并促进神经干细胞分化为神经元和神经胶质细胞,且以神经胶质细胞为主。 Objective To explore the influence of Radix Astragus on proliferation and differentiation of neural stem cells(NSCs) in brain tissue follow cerebral ischemia.Methods Forty-five Wistar rats were randomly divided into sham operation group,control group,and Radix Astragus group.The cerebral ischemia models of rats were established by clamping cerebral middle artery.Radix Astragus(10 ml/kg) was peritoneal injected once 12 h before the operation and once a day post-operation for 1 to 10 d in Radix Astragus group.At the same time,the same volume normal saline was peritoneal injected in sham operation and control groups.The positive cells of Nestin,Nestin combined with stellate fiber acid protein(GFAP) or microtubules related roteins 2(MAP2) in brain tissue were detected by immune-chistochenistry technique at the 3rd,7th and 10th day after the model made.Results(1)Compared with sham operation group,the Nestin positive cells in control group and Radix Astragus group were significantly more(all P〈0.05),and which in Radix Astragus group at each time point were significantly higher than control group(all P〈0.05).(2)The Nestin-GFAP or Nestin+MAP2 positive cells in Radix Astragus group at each time point were significantly more than control group(all P〈0.05).(3)The Nestin-GFAP positive cells were significantly more than the Nestin-GFAP positive cells in control group and Radix Astragus group.Conclusion Radix Astragus can promote brain tissue Nestin expression,and NSCs differentiate into neurons and glial cells,especially glial cells.
