摘要
目的探讨探索学习对局灶性脑梗死大鼠巢蛋白(nestin)及突触素(synaptophysin,SYP)表达的影响。方法健康雄性SD大鼠70只,其中60只经电凝法造成右侧大脑中动脉栓塞(MCAO)模型后,随机分为探索学习组(n=30)和手术对照组(n=30),探索学习组居于探索笼,手术对照组5只一组群居于标准笼;假手术组(n=10)仅开颅不电凝大脑中动脉,居于标准笼。探索学习组和手术对照组分别于术后第1、7、14、28天随机选取5只大鼠处死,假手术组分别于术后第7、28天随机选取5只大鼠处死,即进行巢蛋白及突触素免疫组化染色,测定其在脑梗死灶周围皮质的表达情况。结果探索学习组大鼠巢蛋白阳性神经元数在第7、14天明显多于手术对照组(P<0.01),探索学习组突触素的表达在第14、28天明显多于手术对照组(P<0.01)。结论探索学习能促进梗死灶周围皮质巢蛋白及突触素的表达。
Objective To discuss the effect of learning on nestin and synaptophysin(SYP) expression in rats with unilateral local cerebral infarction.Methods Middle cerebral artery occlusion(MCAO) models,established by electrocoagulation in 60 of 70 healthy SD rats,were randomized into groups learning(n=30) and operation(n=30).The learning group lived in exploring cages,the operation group lived gregariously in standard cages(5 in each cage),sham-operation group(n=10,receiving only craniotomy without electrocoagulation) in standard cages.Five rats from each of groups learning and operation were sacrificed on days 1,7,14,28,and 5 from sham-operation group sacrificed on days 7,28,to perform immunohistochemical staining of nestin and synaptophysin to determine their expressions in peri-ischemic cortex.Results Nestin-positive neurons on days 7,14 and synaptophysin expression on days 14,28 were significantly more in learning group than in control(P〈0.01).Conclusion Learning could enhance nestin and synaptophysin expression in rats with unilateral local cerebral infarction.
