PGE_2和IL1-β对体外培养兔子宫韧带成纤维细胞增殖的影响

Effects of prostaglandin E_2 and interleukin-1β on proliferation of fibroblasts in rabbit uterine ligament cultured in vitro

摘要 目的:探讨前列腺素E2(PGE2)和白细胞介素1-β(IL1-β)在单一和联合应用时,对体外培养兔子宫韧带成纤维细胞增殖的影响。方法:第3代幼兔子宫韧带成纤维细胞,以PGE2(25、50、100、500、1 000ng/ml)、IL1-β(10、20、50、100、200 ng/ml)的单独和联合刺激组为实验组,以常规培养组为对照组,通过MTT法测定细胞增殖效应。结果:单独应用时,100 ng/ml的PGE2和20 ng/ml的IL1-β分别为各自最有效的促增殖浓度(P<0.01);以此浓度联合刺激条件下的细胞增殖活性与对照组及单独刺激相比均有升高(P<0.05)。结论:适当浓度内的PGE2与IL1-β单独或联合使用,能有效促进体外子宫韧带成纤维细胞的增殖,说明PGE2与IL1-β对子宫韧带成纤维细胞的增殖有调节作用。 Objective:To explore the effects of single application or combined application of prostaglandin E2(PGE2) and interleukin-1β(IL-1β) on proliferation of fibroblasts in rabbit uterine ligament cultured in vitro.Methods:The fibroblasts in rabbit uterine ligament of the third generation were obtained,then the fibroblasts treated with single application or combined application of PGE2(25,50,100,500,1 000 ng/ml) and IL-1β(10,20,50,100,200 ng/ml) were selected as experimental group,while the fibroblasts treated with routine method were selected as control group,MTT method was used to detect cell proliferation.Results:PGE2(100 ng/ml) and IL-1β(20 ng/ml) used alone were the most effective doses to promote cell proliferation,respectively(P0.01);when the fibroblasts were treated with PGE2 and IL-1β of the above-mentioned doses,the proliferative activity was higher than those in control group and singe application group(P0.05).Conclusion:Single application or combined application of PGE2 and IL-1β within appropriate doses can promote the proliferation of fibroblasts in uterine ligament in vitro,which indicates that PGE2 and IL1-β can regulate the proliferation of fibroblasts in uterine ligament.
