摘要
目的:探讨丹参滴注液中丹参素钠在Beagle犬体内的药代动力学。方法:健康Beagle犬禁食12 h后,静脉滴注丹参滴注液10 mL.kg-1,血浆样品用乙酸乙酯提取,采用HPLC测定不同时间血药浓度。结果:丹参素钠在0.225~18.000mg.L-1线性关系良好,最低检测限为0.113 mg.L-1,Beagle犬静脉滴注丹参滴注液后的药代动力学参数:tmax为30 min,Cmax为(9.574 2±2.371 5)mg.L-1,t1/2为(19.23±2.97)min,CL为(0.012 7±0.003 0)L.min-1.kg-1,AUC0-tn为(474.954±95.483)mg.min.L-1,AUC0-∞为(482.494±95.353)mg.min.L-1。结论:所建立血药浓度检测方法简便、稳定、可靠,丹参素钠在Beagle犬体内呈单室模型。
Objective:To detect pharmacokinetics of Danshensu Sodium in Danshen dripping solution in Beagles. Method: Dan- shen dripping solution was dripped intravenously into healthy Beagles at a dose of 10 mL · kg-1. Their plasma samples were extracted with acetic ether, the blood concentrations were determined by HPLC method. Result: Danshensu Sodium showed a good linear relation- ship within the range of 0. 225-18.000 mL · kg-1, with the lowest detectable limit of 0. 113mL · kg-1. Its pharmaeokinetic parameters were as follows :tmax was 30 rain, Cm.Was (9. 574 2 ±2. 371 5) mL · kg-1 , t1/2 was ( 19. 23 ±2. 97) min, CL was (0. 012 7 ±0. 003 0) L · min-1· kg-1, AUC0-tn,was (474.954±95.483) mg· min· L-1 and AUC0-∞ was (482.494_±95. 353) mg·min · L-1. Conclu- sion: The accurate, stable and reliable blood concentration method shows a one-compartment mode of Danshensu Sodium in Beagles.
