摘要
合成了2个三元铜(Ⅱ)配合物:[Cu(AMB)(L-Tyr)Cl].1.2H2O(1)和[Cu2(AMB)2(NAA)2Cl2].H2O(2)[AMB=2-氨甲基苯并咪唑,L-Tyr=L-酪氨酸,NAA=萘乙酸根],并通过元素分析、摩尔电导率、IR和UV-Vis对配合物1和2进行了表征。用二倍稀释法测定了配合物1和2的抗菌活性,发现配合物1和2对金黄色葡萄球菌(Staphylococcus aureus,G+)、枯草杆菌(Bacillus subtilis,G+)、沙门氏杆菌(Salmonella,G-)和大肠杆菌(Escherichia coil,G-)具有良好的抑制作用。采用电子吸收光谱、荧光光谱、粘度及琼脂糖凝胶电泳方法研究了配合物1和2与CT-DNA的相互作用。结果表明,2个配合物均以插入方式与CT-DNA作用;在Vc存在下,通过.OH氧化机理切割pBR322 DNA双螺旋结构。2个配合物与CT-DNA相互作用和切割pBR322DNA的强弱均为:配合物2>配合物1。
Two new ternary copper(Ⅱ)complexes:[Cu(AMB)(L-Tyr)Cl]·1.2H2O(1)and[Cu2(AMB)2(NAA)2Cl2]·H2O(2)[AMB=2-aminomethylbenzimidazole,L-Tyr=L-tyrosine,NAA=1-naphthlcetic acid],were synthesized and characterized by elemental analysis,molar conductivity,IR and UV-Vis spectroscope.The complex 1and 2were assayed against gram-positive and gram-negative bacteria by doubling dilutions method,and the interaction with CT-DNA of the two complexes was investigated by electronic absorption spectroscopy,fluorescence spectroscopy,viscosity measurements and agarose gel electrophoresis.The results indicated that the complexes could bind to DNA by intercalative mode,and cleave pBR322 DNA in the presence of vitamin C in the involvement of the hydroxyl radical oxidation mechanism,with the order of the binding ability and cleavage activity of the complexes to DNA:complex 21.
