苦参素注射液对TNBS诱导的结肠炎大鼠结肠黏膜NOD2及IL-6的影响 被引量:2

Influence of oxymatrine injection on expression of NOD2 and IL-6 in colonic intestinal mucosa of rats with experimental colitis

摘要 目的:研究NOD2在实验性大鼠结肠炎发病机制中的作用,并探讨苦参素注射液(OMT)对实验性大鼠结肠炎的治疗作用和机制。方法:将40只SD大鼠随机分为4组:正常对照组、模型组、美沙拉嗪组和OMT组,每组10只。正常对照组未行造模,其余3组均采用三硝基苯磺酸(TNBS)造模。OMT组大鼠给予肌肉注射OMT,美沙拉嗪组大鼠给予美沙拉嗪混悬液灌胃,模型组和正常对照组大鼠均以蒸馏水灌胃,观察实验大鼠结肠黏膜大体形态及组织病理评分,并采用免疫组化法检测大鼠结肠黏膜NOD2蛋白表达,ELISA法检测实验大鼠结肠黏膜IL-6的表达。结果:模型组大鼠结肠黏膜NOD2蛋白、IL-6表达较正常对照组明显升高(P<0.01),美沙拉嗪组、OMT组大鼠结肠黏膜NOD2蛋白、IL-6表达较模型组显著降低(P<0.01)。OMT组大鼠结肠黏膜损伤、结肠黏膜病理组织学评分较模型组明显改善。结论:结肠黏膜NOD2蛋白过度表达、IL-6分泌增多参与了溃疡性结肠炎的发生、发展过程,而OMT可以通过抑制NOD2蛋白过度表达、降低IL-6分泌,起到减轻结肠黏膜炎症,保护结肠黏膜的作用。 Objective: To investigate the role of NOD2 in the pathogenesis of rats′ experimental colitis.And the treatment mechanisms of oxymatrine injection on rats′ experimental colitis were also studied.Methods: Forty Sprague-Dawley(SD) rats were randomly divided into normal control group,model group,SASP group,and OMT group with 10 rats in each group.Except normal control group,colitis model induced by TNBS was established in the remaining three groups.OMT injection was treated with OMT injection,SAP group was given intragastric administration of mesalazine liquids;on the other hand,and the model group and the control group were treated with distilled water.Colon lesion score and histological score of experimental rats were observed.Expression of NOD2 protein in rats colonic mucous was detected by immunohistochemistry.Expression of IL-6 in rats′ colon mucous was detected by ELISA.Results: Compared with normal control group,the expression of NOD2 and IL-6in colonic mucosa of rats in model group were significantly increased(P〈0.01).The expression of NOD2 and IL-6 in colonic mucosa of SASP and OMT group were significantly decreased than that in the model group(P〈0.01).Compared with model group,rat colonic mucosa,and colonic mucosa histological score in OMT group was significantly improved.Conclusion: NOD2 protein is involved with the development of ulcerative colitis.OMT show protection on the colonic mucosa because it can attenuate ulcerative colitis through inhibiting expression of NOD2 and IL-6.
