摘要
目的建立用反相HPLC法测定桦黄中麦角甾醇含量的方法。方法采用CAPCELL PAK C18色谱柱(4.6 mm×150 mm,5μm);甲醇为流动相,流速1.0 ml.min-1;柱温40℃,检测波长283 nm。结果麦角甾醇对照品溶液进样量在0.050 56~1.516 8μg范围内,与峰面积呈良好线性关系,回归方程为Y=1.49×106X-9 245.2,r=0.999 9。样品平均回收率为98.17%,RSD为2.83%,样品粉末加甲醇超声处理60 min,制成供试品溶液。结论该法结果准确、简便可行、分析速度快、分离效果好,适用于桦黄中麦角甾醇的含量测定。
Aim To establish an RP-HPLC method of Ergosterol determination in HuaHuangin.Methods CAPCELL PAKC C18 column(250 mm×4.6 mm,5 μm)was used with a,mobile phase of methanol.The flow rate was 1.0 ml·min-1and the detection wavelength was set at 283 nm with a column temperature of 40℃.Results The linear ranges of the determination of Ergosterol was 0.050 56~1.516 8 μg.The regression equation was Y=1.49×106X-9 245.2,r=0.999 9.The average recovery of Ergosterol was 98.17% and RSD was 2.83%.Powder samples in methanol for 60 minutes ultrasound treatment made the test solution.Conclusion The method is accurate,simple,convenient,fast and the effect of separation is good,so it can be suitable for Ergosterol determination in HuaHuangin.
