摘要
目的:探讨温针治疗肾阳虚证骨关节炎的分子机制,为其疗效提供科学依据。方法:根据肾阳虚证评定量表,选取典型肾阳虚证患者10名给予温针治疗两周,然后以其中疗效最好的4例治疗前后作基因芯片杂交测试,采用SAM基因芯片数据分析软件,筛选显著表达基因作进一步的基因功能分析。结果:4例患者皆为典型肾阳虚证,经温针治疗后效果明显。通过SAM软件筛选到32个显著表达基因,基因功能注释结果提示炎症反应、信号转导类基因显著表达;分子通路主要集中于免疫及信号转导通路。讨论:温针治疗肾阳虚证膝骨关节炎的疗效的分子机制是多方面的,涉及到炎症反应、免疫功能、信号系统等多方面的修复作用。
Objectives: To investigate the molecular mechanisms behind warm needling using Cdna microarray technology,thus providing further scientific evidence for its efficacy.Method: Ten patients with knee OA of kidney-yang deficiency were selected for 2 weeks of warm needling treatment.This treatment involved stimulating the selected acupoints using needles with a burning moxa stick on their handle for 40 minutes per session.Accumulated clinical scores of kidney-yang deficiency were assessed pretreatment and posttreatment using a 40-factor questionnaire of OA with kidney-yang deficiency.Four patients with the best therapeutic efficacy were selected for cDNA microarray testing.The significantly expressed genes were screened through Significance Analysis of Microarray(SAM) for further functionally annotated.Results: 32 significantly expressed genes were screened.GO and PATHWAY annotation results of these genes showed that immune and signal transduction functional genes and pathways significantly expressed.Conclusion: This work demonstrates that the molecular mechanism behind warm needling treatment may be associated with the regulation of immune and signal transduction genes and pathways.
