摘要
以玉米萌动胚为外植体,研究了影响根癌农杆菌介导的玉米NPR1基因遗传转化的若干因素。结果表明,在感染液和共培养基中分别都加入100μmol/L乙酰丁香酮,根癌农杆菌LBA4404的菌液,浸染时间12 h,40 mg/L潮霉素进行转化苗筛选,为转化的适宜条件。对转化苗进行PCR检测,证明外源目的基因已整合到玉米基因组中。
Using germinating embryo of maize as explants,several factors affecting the Agrobacterium-mediated genetic transformation were studied.The results showed that good transformation efficiency could be obtained if the inclusion of acetosyringone in both infection medium and co-cultivation medium was 100 μmol/L;OD600nm of LBA4404 fermenting liquor was 0.6;infection time was 12 h;and co-culture for 16 h at 26 ℃,use 40 mg/L of hygromycin for screening transformed plants.PCR test indicated that the NPR1 gene had been transferred into maize genome.
