摘要
目的研究并建立一种鲜活海产品中重金属汞的快速试纸检测方法。方法对3种汞检测试纸（结晶紫、孔雀石绿和二苯碳酰二肼试纸）进行研究和制备,对试纸的制备条件及显色反应条件进行优化实验。研究并建立海产样品的前处理方法,并主要根据标准色阶的形成情况及添加回收实验结果,对3种试纸的性能进行综合评估和比较,最终选择二苯碳酰二肼试纸作为鲜活海产品中重金属汞的快速检测用试纸。经微波消解处理后的海产样品,在pH 7.5~8.0条件下,可利用二苯碳酰二肼试纸直接进行汞检测。结果二苯碳酰二肼试纸对鲜活海产品汞检测的估计添加回收率为90%~100%,最低检出浓度为1μg.mL-1。实验结果表明,二苯碳酰二肼试纸在制备后至少5~6月内性质稳定,而且,显色反应的日间重复性良好。结论本方法简易、快速、灵敏、可用于鲜活海产品中重金属汞的快速检测。
Objective To study and establish a new approach for rapid-detection of heavy metal mercury in fresh and lively seafood by test paper.Methods Three kinds of mercury-detection test paper（test paper crystal violet,test paper malachite green and test paper diphenylcarbazide） were prepared and studied.Optimization tests were performed both for test paper preparation and coloration reaction.A pre-process method for seafood sample was studied and developed.Comprehensive evaluation and compare were made for the capacity of the three kinds of test paper,mainly based upon the results of spiked recovery tests and standard-coloration-gradients＇ generation.Test paper diphenylcarbazide was eventually selected as the rapid-mercury-detection test paper for seafood sample.Under the condition of a pH of 7.5-8.0,heavy metal mercury in seafood samples could be directly detected by the test paper diphenylcarbazide,after a microwave digestion process.Results The estimated spiked recovery for mercury detection in fresh and lively seafood by test paper diphenylcarbazide was within 90%-100%,and the minimum detect concentration was 1 μg·mL-1.Test paper diphenylcarbazide was proved to be stable in at lest 5-6 months after preparation,and excellent inter-day reproducibility results were obtained for coloration reaction.Conclusion The method is simple,rapid,sensitive and suitable for rapid detection of heavy metal mercury in fresh and lively seafood.
