摘要
目的: 研究髯丝蛛毛苣苔和吊石苣苔的抗菌活性成分。 方法: 以小檗碱为阳性对照,利用纸片扩散法进行抗菌活性研究,液体培养法进行活性成分筛选;采用各种色谱法对高活性部位分离,运用多种波谱技术鉴定结构。 结果: 从髯丝蛛毛苣苔醋酸乙酯部位分离得到5个化合物β-谷甾醇( 1 ),E-3,4-二甲氧基肉桂酸( 2 ),barbinervic acid( 3 ),3 β,19α-二羟基-12-烯-28-乌苏酸 (4),28-O-β-D-glucopyranosyl pomolic acid( 5 )。从吊石苣苔醋酸乙酯部位分离得到3个化合物:5,7-二羟基-6,8,4'-三甲氧基黄酮( 6 ),5,6,4'-三羟基-7,8-二甲氧基黄酮( 7 ),5-羟基-6,8,4'-三甲氧基黄酮-7-O-β-D-葡萄糖苷( 8 )。化合物 3,4,6 对金黄色葡萄球菌(SA)、耐甲氧西林葡萄球菌(MRSA)和β-内酰胺酶阳性的金黄色葡萄球菌(ESBLs-SA) 具有抑制活性。其中化合物 3 (IC50 0.098 g·L^-1)抑制SA活性最好;化合物 4 (IC50 0.130 g·L^-1) 抑制MRSA活性最好;化合物 3 (IC50 0.270 mg·L^-1)抑制ESBLs-SA活性最好。 结论: 化合物 1-5 为首次从植物中分离得到, 7,8 为首次从该科植物中分离得到。
Objective: Antibacteria activity of compounds from Puraboeo rutescens and Lysionotus pauciflorus was assayed. Method: Disc diffusion was used to isolate compounds in vitro and berberine was positive control. The value of IC50 was assayed by the method of liquid culture. All kinds of chromatography were used to isolate the chemical constituent and structure was identified by MS and NMR spectroscopy. Result: Eight compounds were isolated and identified as β -sitosterol (1), E -3, 4-dihydroxy cinnamic acid (2), barbinervic acid (3), 3 β, 19 α -dihydroxy12-en-28-ursolic acid (4), 28- O - β - D -glucopyranosyl pomolic acid (5), 5,7-dihydroxy-6,8,4'-trimethoxy flavone (6), 5, 6, 4'-trihydroxy-7,8-dihydroxy flavone (7), 5-hydroxy-6,8,4'-trimethoxy flavone-7- O - β -D-glucopyranosyl (8). Compound 3, 4 and 6 had activity against SA, MRSA and ESBLs respectively. Compound 3 showed(IC50=0.098 g ·L^-1, IC50=0.27 g ·L^-1) against SA and ESBLs-SA respectively. Compound 4 (IC50 =0.130 g ·L^-1) was best to against MRSA. Conclusion: Compound 1 - 5 were isolated from this plant for the first time. Compound 7 and 8 was isolated from Gesneriaceae for the first time.
