摘要
利用随机扩增DNA多态性(RAPD)技术,对毛乌素沙地天然臭柏种群4种形态变异类型的遗传多样性进行分析。用17个随机引物扩增出201条清晰谱带,其中194条为多态性谱带,并利用POPGENE VER-SION 1.31软件对数据进行处理。结果表明:臭柏4种形态变异类型的遗传多样性较丰富,各类型样本多态位点百分率在79.10%~85.57%之间;基因分化系数为Gst=0.0593,94.07%的遗传变异存在于各变异类型内;聚类分析显示,臭柏枝条的生长特征受遗传控制的可能性较颜色大。
The genetic diversity of four morphological variation types of natural population of Sabina vulgaris in Maowusu Sandy Land was analyzed by use of the Random Amplified Polymorphic DNA(RAPD) technique.201 bands were amplified by seventeen random primers,in which 194 were polymorphic.The data was analyzed with the POPGENE VERSION 1.31 software.The results showed that the genetic diversity of four morphological variation types of Sabina vulgaris was abundant.The percentage of polymorphic loci(PPB) for all types varied from 79.10% to 85.57%.Gst=0.0593,94.07 percent of genetic differentiation were within all variation types.The cluster analysis demonstrated that the genetic charged by growth characteristics of breach was more possible than color.
