摘要
目的:观察益肾胶囊对大鼠肾小球系膜细胞(glomerular mesangial cell,GMC)增殖、凋亡及TGF-β1 mRNA表达的影响。方法:以体外培养肾小球系膜细胞为研究对象,应用血清药理学方法,分别采用甲基噻唑基四唑(methyl thiazolyl tetrazolium,MTT)比色法、荧光倒置显微镜及RT-PCR技术检测系膜细胞增殖、凋亡及TGF-β1 mRNA表达情况。结果:与正常对照组比较,其他各组MC均有不同程度的增殖、凋亡及TGF-β1 mRNA表达上调(P<0.05或P<0.01);与脂多糖(lipopolysaccharides,LPS)组比较,益肾胶囊可明显抑制MC增殖及TGF-β1 mRNA表达,并诱导MC凋亡(P<0.01)。结论:益肾胶囊抑制MC增殖并诱导其凋亡可能与下调TGF-β1 mRNA表达有关。
Objective：To investigate the effects of yishen capsule on the proliferation,apoptosis and expression of TGF-β1 mRNA of rat glomerular mesangial cell （GMC）.Methods：Utilizing the method of serum pharmacology to study the cultured GMC,and the proliferation,apoptosis and expression of TGF-β1 mRNA of GMC were detected by methyl thiazolyl tetrazolium （MTT）,fluorescence inverted microscope and the technique method respectively.Results：Compared with control group,the proliferation,apoptosis and TGF-β1 mRNA expression of GMC of three other groups were significantly different （P0.05 or P0.01）,compared with LPS group,Yishen Capsule could inhibit the proliferation,expression of TGF-β1 mRNA and promote the apoptosis of GMC （P0.01）.Conclusion：Yishen Capsule could inhibit the proliferation and promote the apoptosis of GMC maybe correlate with decrease the expression of TGF-β1 mRNA.
