顺式氯氰菊酯离体拟雌激素活性评价及机制初探 被引量:1

摘要 目的研究顺式氯氰菊酯对人类乳腺癌MCF-7细胞增殖及对其雌激素受体αmRNA表达的影响。方法顺式氯氰菊酯分为1.0×10-7、1.0×10-8、1.0×10-9mol/L 3个剂量组,阳性对照β-雌二醇分为1.0×10-7、1.0×10-9、1.0×10-11mol/L剂量组,以及溶剂对照和空白对照组。MCF-7细胞按以上分组于96孔板培养5 d后测吸光度值,计算细胞增殖率;相同分组及方法培养MCF-7细胞后提取RNA,逆转录,应用实时定量聚合酶链反应确定雌激素受体αmR-NA的表达量。结果不同浓度的顺式氯氰菊酯与溶剂对照组比较,细胞增殖率均升高(P<0.01)。1.0×10-7、1.0×10-8、1.0×10-9mol/L浓度顺式氯氰菊酯处理MCF-7细胞的雌激素受体αmRNA表达水平上调,分别为溶剂对照组的1.61、1.45和1.39倍。结论顺式氯氰菊酯可显著促进MCF-7细胞的生长,表现出拟雌激素样作用,并使MCF-7细胞雌激素受体αmRNA表达上调。 Objective To study the effects of alpha-cypermethrin on cell proliferation and ERα mRNA expression in human breast cancer cell line(MCF-7).Methods MCF-7 cells were cultivated for 5 days in 96-well plates with alpha-cypermethrin(3 groups,1.0×10-7,1.0×10-8 and 1.0×10-9 mol/L),β-estradiol(3 groups,1.0×10-7,1.0×10-9 and 1.0×10-11 mol/L).A solvent control and a blank control were also applied.Absorbance values were measured and the proliferation rates were calculated;MCF-7 cells were also cultivated by the same method.Their RNA were extracted and reversely transcripted,then the mRNA expression of ERα was observed by real-time PCR.Results Compared to the solvent control,the proliferation rates in all alpha-cypermethrin groups were significantly increased(P0.01).ERα mRNA expression levels in alpha-cypermethrin groups of 1.0×10-7,1.0×10-8 and 1.0×10-9 mol/L were 1.61,1.45 and 1.39 fold higher than those in the solvent control separately.Conclusion Alpha-cypermethrin can significantly promote MCF-7 cell proliferation,display estrogencity-like activity,and up-regulate the ERα mRNA expression.
