摘要
神经生长因子(nerve growth factor,NGF)是最早发现的神经营养因子家族成员,它主要通过细胞表面受体酪氨酸蛋白激酶A(tyrosine kinase A,TrkA)来介导复杂的生物学效应。TGF-β(transformation growth factor-β)是一个包含众多成员的细胞因子超家族,它们通过Smad介导的经典信号通路或者不依赖于Smad蛋白的信号通路发挥功能。有报导称,在鼠嗜铬细胞瘤(PC12)细胞中,NGF可以激活Smad信号通路,但其具体分子机制依然不清楚。为了进一步了解NGF介导的信号与TGF-β信号之间的串话(crosstalk),采用GST-pulldown和免疫共沉淀-免疫杂交的方法,对NGF受体和TGF-β受体之间的相互作用进行了研究,发现NGF受体TrkA与TGF-β超家族的各种I型受体之间均存在相互作用;同时,TrkA也能与TGF-β超家族的Ⅱ型受体TβRII及ActRII结合,但并未检测到Tr-kA与另一个Ⅱ型受体BMPRII之间的相互作用。
The nerve growth factor(NGF) is a member of the neurotrophin family and was the first neurotrophic factor has been isolated.It mediates many complex biological processes through the specific receptor,tyrosine protein kinase A(TrkA).The TGF-β superfamily includes many cytokines,which transduce their signal through Smad-dependent or-independent pathways.It has been reported that in rat pheochromocytoma cells(PC12),NGF stimulation resulted in activation of Smad cascade,while the mechanism of this process is still unknown.To obtain better understanding of the crosstalk between NGF and TGF-β signaling,the present study detected the interaction between NGF receptor TrkA and TGF-β superfamily receptors by GST-pulldown and immunoprecipitation-immunoblotting assays.It found that all TGF-β superfamily type I receptors could bind to TrkA,and TGF-β superfamily type II receptors TβRII and ActRII could also interact with TrkA.But no interaction was detected between BMP receptor II and TrkA.
