绿色木霉C-08产纤维素酶的固态发酵条件优化 被引量:6

摘要 以稻草粉与麦麸为主料,对影响绿色木霉(Trichoderma viride)C-08固态发酵生产纤维素酶、半纤维素酶的营养因素和环境条件进行了研究。结果表明:秸秆粉和麸皮为2∶4最适合产酶;铵态氮适合C-08产CMCase,NO3-利于产Xylanase,其中NH4NO3最好;料水比1∶2,适合C-08产CMCase,1∶2.5适合产Xylanase;稻草粉粒度40~100目时,适合C-08产CMCase,此范围外适合产Xylanase;pH值为4时,适合C-08产酶;孢子接种量为7.5×107孢子.mL-1时,适合C-08产酶;低于28℃时适合C-08产酶;最适发酵时间为72h。该菌株在以上条件下产酶效果较好,为实际应用奠定了基础。 Trichoderma viride C-08 can very efficiently produce cellulase and xylanase.The optimal conditions of the solid-state fermentation for Trichoderma viride to produce cellulase and xylanase were studied.The results showed that optimal solid culture medium had ratios of straw to bran 2:4.NH4+ source was good for C-08 to produce cellulose and NO3-source was good for C-08 to produce xylanase,so NH4NO3 source was the best.The ratio of stuff to water 1:2 was good for C-08 CMCase production,and 1:2.5 for Xylanase production.The optimal rice straw granularity 150-425μm was suitable for C-08 CMCase production.Granularity below 150μm and above 425μm was best for xylanase production.The optimal pH for C-08 fermentation was 4.The optimal conditions for cellulase and xylanase production were as follows: inoculum size 7.5×107,incubation temperature lower than 28℃,fermentation period 72h.
