扇贝多肽抑制中波紫外线导致的人皮肤成纤维细胞周期阻滞 被引量:4

Polypeptides from Chlamys farreri(PCF) inhibit human fibroblast cell cycle turbulence induced by UVB irradiation

摘要 目的探讨扇贝多肽(PCF)对中波紫外线(UVB)辐射导致的细胞周期阻滞的作用。方法应用流式细胞术检测扇贝多肽对细胞周期的影响;蛋白质印迹检测p53和p21Cip1的变化;2,7-二氯氢化荧光素二酯(DCFH-DA)探针检测活性氧(ROS)的变化。结果 UVB辐射导致人皮肤成纤维细胞周期发生G1期阻滞,扇贝多肽缓解了UVB辐射诱导的细胞周期阻滞,抑制了细胞的p53及p21Cip1蛋白表达,清除了细胞中的ROS。结论扇贝多肽抑制了UVB辐射引起的人皮肤成纤维细胞G1期阻滞,其保护作用是通过抑制ROS-p53-p21Cip1途径实现的。 Objective To investigate the effect of polypeptides from Chlamys farreri on UVB-induced human fibroblast cell cycle arrest.Methods Flow cytometry was preformed to analyze the human fibroblast cell cycle;p53 and p21Cip1 protein were detected by Western Blot;ROS was determined by DCFH-DA probe.Results UVB irradiation resulted in G1 arrest in human fibroblast and PCF pretreatment attenuated the cell cycle arrest.P53 protein was decreased in the present of PCF and followed by a p21Cip1 protein-depression.ROS was scavenged by PCF pretreatment.Conclusion PCF attenuated the cell cycle arrest induced by UVB irradiation in human fibroblast.The protective effect was via the inhibition of ROS-p53-p21Cip1 pathway.
