黄芪多糖对糖尿病大鼠神经肽Y及其Y_2受体表达的影响 被引量:8

Effect of Astragalus Polysaccharides on Neuropeptide Y and Its Receptor Y_2 Expression in Streptozotocin-induced Diabetic Rats

摘要 目的:探讨黄芪多糖(APS)对糖尿病大鼠肾脏神经肽Y(NPY)及其Y2受体(Y2R)表达的影响。方法:采用腹腔注射链脲佐菌素(STZ)制备糖尿病大鼠动物模型,随机分为对照组、糖尿病组和黄芪多糖干预组;采用放射免疫法检测血浆和肾皮质NPY水平,实时荧光定量PCR方法检测肾皮质NPYmRNA的表达,免疫组织化学方法检测肾皮质Y2R蛋白表达,同时检测大鼠的血糖、尿白蛋白排泄率(UAER)、肾重/体重。结果:①APS干预能显著削弱糖尿病大鼠血糖、UAER和肾重/体重的增加;②APS干预后显著降低糖尿病大鼠血浆和肾皮质NPY水平(P<0.01);③APS干预组大鼠肾皮质NPYmRNA和Y2R蛋白表达显著低于糖尿病组(P<0.05)。结论:APS对糖尿病肾病的保护作用机制可能与下调肾脏NPY及其Y2R的表达有关。 Objective:To investigate the effects of astragalus polysaccharides on the expression of neuropeptide Y(NPY)and its receptor Y2(Y2R)in the kidney of diabetic rat.Methods:The male wistar rats were randomly allocated to three groups:normal control group,STZ-induced diabetic rats group and diabetic rats treated with Astragalus Polysaccharides(APS)group(400mg·kg-1·d-1,i.p.28d),After 28 days,the rats were sacrificed and the various components were determined.The level of NPY in plasma and renal cortex,the urine albumin excretion ratio(UAER)were measured by radioimmunoassay(RIA).RT-PCR and Immunohistochemistry(IHC)and computer software for image analysis to evaluate the expression of NPY mRNA and Y2R.Blood glucose,body weight,kidney weight were measured.Results:①Compared with diabetes mellitus group,the UAER,blood glucose and kidney weight/body weight,of diabetic rats were ameliorated by APS treatment.②Compared with diabetes mellitus group,the plasma and renal cortical NPY concentrations were significantly decreased(P0.01)in APS treatment group.③The expressions of NPY mRNA and Y2R protein were significantly decreased in the renal cortex of diabetic rats treated with Astragalus Polysaccharides as compared with diabetic rats(both P0.05).Conclusion:These results suggest that APS exerts its therapeutic effects on diabetes mellitus,which may be related to the down regulation expression of NPY mRNA and Y2R in the kidney of diabetic rats.
