摘要
目的探讨超微通阳调心方对大鼠扩张型心肌病模型血浆脑利钠肽(BNP)及心肌细胞凋亡的影响。方法将60只大鼠随机分成5组,分别为正常对照组、扩张型心肌病(DCM)模型组、超微饮片1/3剂量组、超微饮片等剂量组及普通饮片等剂量组。除正常组外,其余4组均以阿霉素腹腔注射建立DCM模型,BNP指标测定评价心功能改变,Bax、Bcl-2指标测定心肌细胞凋亡。结果普通饮片等剂量组及超微饮片等剂量组均可提高Bcl-2,降低Bax,与DCM模型组比较差异有统计学意义(P<0.05),普通饮片与超微饮片等剂量组比较(P>0.05),超微饮片与模型组比较(P<0.05);BNP水平DCM模型组较正常对照组明显增大(P<0.01),超微饮片等剂量组及普通饮片等剂量组较DCM模型组明显减小(P<0.05)。提示普通饮片及超微饮片均可改善心功能,降低BNP,抗心肌细胞凋亡。结论通阳调心方能改善阿霉素所致大鼠扩张型心肌病模型的心功能,降低神经体液因子BNP水平,并有抗心肌细胞凋亡的效应。
Objective To explore the effects of ultramicro Tongyang Tiaoxin decoction(UTTD) on cardiomyocyte apoptosis and BNP in rats with dilated cardiomyopathy(DCM).Method Sixty rats were divided randomly into five groups,as normal group,DCM group,1/3 UTTD dose group,normal UTTD dose group and traditional decoction group.Except for the normal group,all the rats were given Doxorubicin by intravenous injection twice per week in 8 weeks to induce the DCM models.The changes of cardiac function were measured with BNP and the cardiomyocyte apoptosis were detected with Bax and Bcl-2.Result Bcl-2 increased and Bax decreased in both traditional decoction group and UTTD dose group,but the difference was not signigieant(P0.05).The BNP level in DCM group was increased than that in normal group (P0.05),while in normal UTTD dose group and traditional decoction group were decreased than that in DCM group(P0.05).Conclusion Ultramicro Tiaoxin decoction can improve cardiac function,lower BNP levels of neurohumoral factors and be acted to resist the cardiomyocyte apoptosis.
