Anti-tumor activity of petroleum extract from Herba Scutellariae Barbatae and determination of its components

摘要 目的:研究半枝莲石油醚提取物体外抗肿瘤活性,并测定其成分。方法:甲基偶氮唑盐类(MTT)法研究该提取物对MGC-803细胞、K562细胞和HepG-2细胞的抑制作用;并用气相色谱-质谱联用(GC-MS)法测定其成分。结果:该部位对3种肿瘤细胞IC50分别为18.4,21.7,56.2mg·mL-1;该提取物主要含有不饱和脂肪酸、酯类及甾醇等成分,其中棕榈酸乙酯含量最高(23.53%)。结论:该提取物对3种肿瘤细胞均有抑制作用;半枝莲石油醚提取物中的主要抗肿瘤活性成分为不饱和脂肪酸和豆甾醇。 OBJECTIVE To investigate anti-tumor effects of petroleum extract from Scutellaria Barbata Don, and determine its chemical constituents. METHODS This extract was tested for anti tumor activity in vitro with three kinds of tumor cells (MC, C-803 cells, K562 cells, HepC-2 cells) by MTT. Components were determined by GC-MS. RESULTS The extract from Scutellaria Barbara hibited the proliferation of these three kinds of tumor cells, and ICsa values for MGC-803 cells, K562 cells and HepG-2 cells were 18.4μg·L^-1 , 21.7μg·L^-1 and 56. 2μg·L^-1, respectively. Components were identified as following: unsaturated fatty acid, esters, sterol and others, and hexadecanoic acid ethyl ester is the main component (23.53%). CONCLUSION The extract has from Scutellaria Barbata inhibition effect on the proliferation of three tumor cells in vitro. From the GC MS analysis results, it suggests that the main antitumor active constituents could be unsaturated fatty acid and stigma-sterol.
