摘要
目的脂肪细胞分化的调控与肥胖及胰岛素抵抗的发病机制有密切关系。文中研究蒲黄总黄酮(pollen typhae total flavone,PTF)对3T3-L1前脂肪细胞增殖分化的影响,并通过相关基因的表达分析其可能机制。方法通过XTT法检测各剂量PTF干预的前脂肪细胞增殖的变化,对PTF干预分化的细胞进行油红O染色并通过比色定量分析脂肪细胞分化程度。RT-PCR检测脂肪细胞分化相关基因过氧化物酶体增殖物激活受体γ(peroxisome proliferator activated receptorγ,PPARγ)、CAAT/增强子结合蛋白α(CAAT/enhancer binding proteinα,C/EBPα)mRNA的表达。结果 PTF明显促进前脂肪细胞的增殖,在0.2 g/L时可明显促进前脂肪细胞的分化,但分化程度低于罗格列酮组。PTF可提高PPARγ和C/EBPαmRNA表达,与对照组比较差异有显著性意义(P<0.01)。结论 PTF促进前脂肪细胞增殖分化水平,其机制可能与提高PPARγ和C/EBPα的mRNA表达有关。
Objective The regulation of adipocyte differentiation is closely related with the pathogenesis of obesity and insulin resistance.This study was to investigate the effects of pollen typhae total flavone(PTF) on the proliferation and differentiation of 3T3-L1 pre-adipocytes and its possible mechanism by measuring the related gene mRNA expression.Methods The proliferation of pre-adipocytes treated with PTF was measured by XTT and stained in the differentiation phase by oil red O,and their differentiation was quantitatively analyzed by spectrophotography.The pre-adipocyte differentiation related expressions of PPARγ and C/EBPα mRNA were determined by real-time PCR(RT-PCR).Results PTF obviously promoted the proliferation and,at 0.2 g/L,the differentiation of pre-adipocytes.It also up-regulated the expressions of PPARγ and C/EBPα mRNA,with significant differences from the control group(P 0.01).Conclusion PTF can promote the proliferation and differentiation of 3T3-L1 pre-adipocytes by up-regulating the expressions of PPARγ and C/EBPα mRNA.
