摘要 目的近来研究显示,人眼眶脂肪组织中的前脂肪细胞可能参与多种疾病的发病过程。文中建立人眼眶前脂肪细胞原代培养模式,对其进行培养、诱导分化及鉴定,以便深入了解人眼眶前脂肪细胞的生物学特性。方法从18~56岁成人眼眶脂肪组织中分别以组织块法及酶消化法分离得到前脂肪细胞,进行形态学观察,成脂诱导分化,并对其进行鉴定。结果 2种方法均可获得前脂肪细胞,细胞为长梭形,呈"S"形生长,可成脂分化,且油红O染色阳性。结论从人眼眶脂肪组织中可分离出纯度高、增殖快、成脂分化率高的前脂肪细胞。 Objective Recent studies have shown that human orbital preadipocytes play an important part in many diseases.Our research is to establish a primary human orbital preadipocyte culturing method for the culture,induction,differentiation and identification of human orbital preadipocytes and gain a deeper insight into their biological traits.Methods We used the methods of tissue culture and enzyme digestion to obtain preadipocytes from the orbital adipose tissue of humans aged 18-56 years,and observed their morphology,proliferation and differentiation,followed by identification.Results Preadipocytes were successfully obtained by both the methods of tissue culture and enzyme digestion.Enzyme-digesting methods got purer and more preadipocytes,which were fusiform and grew in an 'S' shape,with lipid droplets,positive on oil red O staining.Conclusion Human orbital preadipocytes can be differentiated into mature lipid-filled adiocytes.
