摘要
目的:观察TGF-β1在阻塞性黄疸肝组织中的表达,并初步探讨其作用。方法:运用Wistar大鼠建立阻寨性黄疸模型,观察其肝脏病理变化及血清胆红素的变化;运用免疫组织化学方法检测肝脏组织中TCF-β1的表达;并运用原位末端标记方法检测肝细胞凋亡。结果:Wistar大鼠结扎胆总管后,胆红素急剧升高,术后1周可见肝脏表达TCF-β1,肝细胞出现大量的凋亡,纤维组织增生,术后2周TGF-β1表达及细胞凋亡达到高峰,纤维组织大量增生,术后3周纤维组织占据肝脏组织大部分,细胞凋亡相对减少,仍可见大量的TGF-β1表达。结论:在阻塞性黄疸中,肝脏表达大量的TGF-β1,与肝细胞凋亡、纤维化及机体免疫功能抑制密切相关。
Objective: To observe the expression of transforming growth factor-β_1(TGF-β_1 ) in heqatictissue of obstructive jaundice, and to explore is prelinunary effect.Methods: Hepatopathology and serum bilirubin were observed by establishing obstructive jaundice model with Wistar rats . The experession of TGF-β_1 inhepatic tissue was detecled with immunohositochemistry; and hepatocellular apoptosis with in situ end-labeling. Results: For Wistar rats after ligation of commonduct, bilirubin increased accordingly. At the end of oneweek after ligation, We could see the expressison of TCF-β1 in liver, which showed a greal deal of cellutar apoptosis and fibroplasia. After two weeks, the expression of TCF-β1 and apoptosis reachedto a peak , and therewere intense fibrous tissue proliferations. At the end of three weeks fibrous tissue occupied most of the liver andapoptosis began to decline. But we could still see a large amount of TGF-β1, expression. Conclusion: Duringthe time of obstructive jaundice, liver expressed a great deal of TGF-β1, which was closely related to hepatocellalar apoptosis, fibrosis and immuse function inhibition of the body.
