暗纹东方鲀线粒体ATPase8和ATPase6基因的克隆与序列分析 被引量:5

Cloning and sequence analysis of the ATP Synthase Fo Subunits (ATPase8 and ATPase6) genes of mitochondrial DNA genome from Takifugu fasciatus

摘要 克隆并测定了暗纹东方鲀(Takifugu fasciatus)线粒体ATP合酶Fo亚基8(ATPase8)和亚基6(ATPase6)的序列,并对其进行了初步分析。结果表明:PCR扩增产物的总长度为923 bp,其中842 bp为ATP8和ATP6基因的编码区。ATP8基因长168 bp,编码55个氨基酸残基的蛋白质,其蛋白分子质量为6.8 KD,等电点为7.85;ATP6基因长684 bp,编码227个氨基酸残基的蛋白质,其蛋白分子质量为24.9 KD,等电点为9.16。暗纹东方鲀ATP合酶Fo亚基8和亚基6与其他已报道的部分东方鲀属鱼类具有很高的同源性。通过探讨ATP合酶Fo亚基所含的信息量,发现相对于其他线粒体基因,ATPase8和ATPase6基因所含鉴别信息较少。 Mitochondfial ATP synthase Fo subuitit 8(ATPase8) and subunit 6 (ATPase6) genes in liver tissue of Takifugu fasciatus were cloned, sequenced and analyzed preliminarily with primers designed according to ATP8 and ATP6 genes of Takifugu rubripes. The results show that the full length of the PCR product is 923 bp which contains 842 bp ORF encoding the ATP8 and ATP6 genes with a 10 bp overlap between them, The ATP8 sequence is 168 bp encoding a protein with 55 amino acid residues whose molecular weight is 6.8 KD and isoelectric point is 7.85. The ATP6 gene is 684 bp encoding a protein with 227 amino acid residues whose molecular weight is 24.9 KD and isoelectrie point is 9.16. Both of them are highly homologous to those of other fishes reported. Phylogenetic tree based on nucleotide sequences constructed by NJ and MP method using MEGA3.1 manifests that the Takifugu fasciatus have closer relationship to the Takifugu rubripes, and that the ATP8 and ATP6 genes have less phylogenetie information than other mitochondrial genes.
