摘要
目的:通过补肾中药诱导骨髓间充质干细胞(MSCs)定向分化的研究,阐述"肾主骨、生髓"理论。方法:中药药液灌胃SD大鼠4d后经腹主动脉采血制备含药血清。将第4代MSCs以5×106接种于2块6孔板,每板6孔,分为空白组、巴戟天组、杜仲组、补骨脂组、肉苁蓉组、淫羊藿组。细胞长至80%时,各中药组加入含药血清L-DMEM培养基,空白组原培养基继续培养;诱导第10天取出1块6孔板做ALP染色;另一块第20天做6孔板茜素红染色。结果:巴戟天组、杜仲组、肉苁蓉组、淫羊藿组于第10天ALP染色阳性,第14天左右起可见白色钙结节,第20天茜素红染色阳性。结论:以"肾主骨、生髓"理论指导,补肾中药能诱导骨髓间充质干细胞向成骨细胞分化。
Objective:To explain the theory of kidney determining the condition of bone and marrow by directional differentiation of bone marrow mesenchymal stem cells (MSCs) with Chinese drug which can nourish the kidney. Methods:Serum containing Chinese drug was abstracted from rats' blood after intragastric administration with Chinese liquid drug for 4 days. MSCs with 5×10^6 per milliliter were cultured in two six-well cell culture plates and divided into control group,Bajitian (Radix Morindae Officinalis) group,Buguzhi (Psoralea corylifolia L) group,and. All Chinese drug groups were added blood serum containing Chinese drug while control group was added the previous medium. Ten days later,one cell culture plate was colored by AKP staining solution,and twenty days later,the other one was colored by alizarin Bordeaux. Results:ALP staining showed positive results on the 10th day in Bajitian (Radix Morindae Officinalis),Duzhong (Cortex Eucommiae),Roucongrong (Herba Cistanches) and Yinyanghuo (Herba Epimedii) groups. From the 14th day on,white calcium nodus could be seen,and could be colored by alizarin Bordeaux twenty days later. Conclusion:Chinese drugs which can nourish the kidney are able to induce MSCs directional differentiation by the theory of kidney determining the condition of bone and marrow.
