摘要
目的建立视网膜母细胞瘤裸鼠玻璃体腔移植瘤模型并观察其生长状态,为开展肿瘤体内实验做好前期工作。方法培养视网膜母细胞瘤Y79细胞,调整细胞浓度为(4.5-5.5)×107/ml,注射至裸鼠玻璃体腔内(5μl/只),观察肿瘤的生长状态。结果注射早期玻璃体腔内浑浊明显,而后依次出现肿瘤团块;角膜混浊;眼内结构破坏;眼球突出于眼外。摘除的肿瘤组织HE染色结果显示瘤细胞大小不等,胞浆少,核大,核分裂相常见。结论利用Y79细胞系可以建立良好的视网膜母细胞瘤玻璃体腔移植瘤模型,为人们探索视网膜母细胞瘤的发展规律以及治疗方法提供了有用的工具。
Objective Methods Y79 cells were cultured and injected into vitreous cavity of the nude mice. Each animal was injected 5μl at a concentration of (4.5 - 5.5)× 107/ml. The growth of tumors was observed. Results The progression of tumor growth was as following: vitreous cavity becoming not clear, tumor clot appearing, corneal opacity, structure of ocular being destroyed, eyeball extnlding from orbital cavity. HE staining of tumor enucleated showed that the tumor cells differed in size, the nucleus was large, the cytoplasm was lack and the nuclear division was common. Conclusion The Y79 retinoblastoma model can be established successfully in nude mice vitreous cavity. It would be a useful tool to investigate the progression of retinoblastoma and method of tumor therapy.
