摘要
目的探讨原发性肝细胞癌(肝癌)患者外周血调节性T细胞亚群变化及其临床意义。方法应用流式细胞仪检测34例肝癌患者及30例同龄健康人群外周血单个核细胞(PBMC)中CD4+CD25high T细胞占CD4+T细胞的比例;MTT法用于检测部分病人外周血PBMC的增殖情况。结果肝癌组CD4+CD25high/CD4+的比率较健康对照组显著增加(P<0.01);肝癌Ⅲ期患者CD4+CD25high/CD4+T比例高于Ⅰ、Ⅱ期(P<0.01);肝癌组PBMC增殖水平较健康对照组明显下降(P<0.01)。结论肝癌患者CD4+CD25high调节性T细胞比例显著升高,外周血PBMC增殖水平较健康人群明显下降,可能与肝癌导致机体免疫功能异常有关。
Objective To investigate the frequency of CD4^CD25^+ regulatory T cells and its clinical significance in the peripheral blood of the patients with hepatocellular carcinoma ( HCC ). Methods Flow cytometer was used to analyze the frequency of regulatory T cell (Treg) in peripheral blood mononuclear cells (PBMC) from both 34 patients with HCC and 30 normal donors. The MTT method was used to observe changes in PBMC proliferative function. Results The frequency of CD4^+CD25^high regulatory T cells in the peripheral blood of hepatocellular carcinoma patients was higher than that in the control group (P 〈 0.01 ). The levels of Tregs in peripheral blood were correlated with disease progression in HCC patients. The proliferative function of PBMC in patients with hepatocellular carcinoma was decreased (P 〈 0.01 ). Conclusion The decreased proliferative function of PBMC and the increased frequency of regulatory T cells in patients with hepatocellular carcinoma may be one of the mechanisms that immune function was impaired in patients with HCC.
