曼宋酮E对白血病K562细胞增殖和凋亡的影响 被引量:2

摘要 目的观察曼宋酮E对K562细胞增殖的影响,探讨其诱导凋亡作用及可能机制。方法台盼蓝拒染实验检测曼宋酮E对K562细胞的生长抑制作用;荧光显微镜观察细胞核形态的变化;流式细胞仪测定细胞凋亡率;免疫印迹法测定caspase-3、caspase-8、caspase-9及PARP的活性及Bcl-2、Bax和Bcl-XL的表达。结果曼宋酮E抑制K562细胞增殖呈时间和浓度相关性;12.5μg/mL的曼宋酮E处理K562细胞0、12、24、48h细胞凋亡率分别为(2.1±0.8)%、(3.2±1.6)%、(15.3±8.9)%、(25.1±11.4)%;在曼宋酮E诱导K562细胞凋亡过程中,caspase-3和caspase-8以及PARP出现活化断裂,casepase-9表达无明显变化;Bcl-2家族蛋白Bcl-2、Bax和Bcl-L表达无显著改变。结论曼宋酮E可抑制诱导K562细胞增殖,并通过caspase-8途径介导凋亡。
