摘要
目的:通过观察实验性精索静脉曲张大鼠睾丸生精细胞凋亡指数(AI)、睾丸组织Caspase-3,Hsp60表达变化,探讨实验性精索静脉曲张大鼠睾丸生精细胞凋亡状况及通精灵对精索静脉曲张大鼠睾丸生精功能的保护作用。方法:选择成年清洁级雄性W istar大鼠50只,按Saypol法制作左精索静脉曲张大鼠模型,随机分为假手术组10只、模型组10只、通精灵低剂量组10只、通精灵中剂量组10只、通精灵高剂量组10只。造模1个月后,通精灵各组予以不同浓度的通精灵水煎剂,通精灵各组均每日1次灌胃。假手术组和模型组予以等量的生理盐水灌胃,每日1次,连续2个月。各组实验大鼠普食喂养2个月后,全部处死,取出睾丸,用原位末端脱氧核苷酸转移酶标记法(TUNEL)检测睾丸生精细胞凋亡,免疫组化法检测睾丸组织Caspase-3、Hsp60表达。结果:①模型组大鼠左睾丸凋亡指数明显高于假手术组及通精灵各组(P<0.01),通精灵各组左睾丸凋亡指数与模型组比较,差异有显著性(P<0.05)。②模型组大鼠睾丸组织Caspase-3、Hsp60蛋白表达上升;与假手术组比较,差异有显著性(P<0.01,P<0.05),通精灵中、高剂量组Caspase-3、Hsp60蛋白表达显著下降,与模型组比较,差异有显著性(P<0.05)。结论:通精灵能降低实验性精索静脉曲张大鼠睾丸生精细胞凋亡,减轻实验性精索静脉曲张大鼠睾丸组织的病理损害;通精灵可下调Caspase-3、Hsp60表达,这可能是通精灵抑制生精细胞凋亡,改善精索静脉曲张不育症患者生精功能机制之一。
Objective：To investigate the apoptosis of spermatogenic cell and the protective effect of Tongjingling decoction on spermatogenic function in testis of rats with varicocele by observing the apoptosis index in spermatogenic cells,the expression of Caspase-3 and Hsp60 gene in the testis.Methods：Referring to Saypol＇s method,experimental left varicocele was induced by partial ligation of the left renal vein in 50 Wistar rats,which were randomly divided into 5 groups as follows：sham operation group,model group,Tongjingling low dose group,Tongjingling middle dose group,Tongjingling high dose group,with 10 cases in each group.After one month models were made sucessfully,medicine was given for two months.Tongjingling groups were given Tongjingling decoction with different concentrations once time every day,sham operation group and model group were given the same dosage of normal saline one time every day.Two months later the rats were killed and the testes were selected,the apoptosis cells were detected by in situterminal deoxynucleotityl transferase-mediated dTUP nick end lableling（TUNEL）technique,measure the expression of caspase-3,hsp60 gene with the method of immunohistochemistry.Results： ①Apoptosis index in spermatogenic cells of mode group were distinctly higher than that in the sham operation group and Tongjingling groups（P〈0.01）,the contrast between Tongjingling groups and model group was manifest（P〈0.05）.②The exprssion of Caspase-3 and Hsp60 markedly increased;compared with the sham operation group,the difference was significant（P〈0.01,P〈0.05）.The expression of Caspase-3 and Hsp60 markedly decreased,compared with the model group,the difference was significant（P〈0.05）.Conclusion：Tongjingling have negative effect on the apoptosis of spermatogenic cell,can relive the pathological harm in testis of experimental rats with varicocele;Tongjingling decrease the expression of Caspase-3 and Hsp60 gene.This may be one of the mechanism of Tongjingling resisting the apoptosis of spermatoblast cell induced by varicocele and improving the capability of bearing.
