摘要
目的:探讨α-生育酚对精索静脉曲张(VC)大鼠睾丸生殖细胞损害的保护作用。方法:采用成年雄性Wistar大鼠10只复制精索静脉曲张疾病模型,4周后开始每日肌注α-生育酚0.5mg/(100g体重),共4周(生育酚组)。同时设手术组及假手术组进行对照。8周后用原位末端脱氧核苷酸转移酶标记法(TUNEL)检测睾丸生殖细胞凋亡指数(AI)。结果:生育酚组动物左侧睾丸重量显著高于手术组(P<0.05),低于假手术组(P<0.05);生育酚组细胞凋亡率[左侧(5.34±1.28)%,右侧(5.87±1.25)%]高于假手术组[左侧(4.72±1.02)%、右侧(4.88±1.28)%](P<0.05),而显著低于手术组[左侧(9.05±2.51)%,右侧(7.22±1.35)%](P<0.05)。结论:α-生育酚能降低精索静脉曲张大鼠生殖细胞的凋亡指数,延缓睾丸细胞损害的病程,对精索静脉曲张导致的睾丸损害具有一定的保护作用。
Objective:To study the effects of α-tocopherol on spermategenic cellular apoptosis in experimental varicocele in rats. Methods:Thirty adult male Wistar rats were randomized into 3 groups( n =10): treated group (TG), operation group (OG). and sham operation group (SOG). Rats in treated group were injected intramusculously α-tocopherol (0.5 mg/100 g body-weight, one time daily for 4 weeks) after varicocele being induced by partial ligation of the left renal vein for 4 weeks. All the rats were breed in the similar condition for eight weeks. The apoptosis cells were detected by in situ terminal deoxynucleotityl transt'erase mediated dTUP nick end lableling (TUNEL). Results:The left vena spermatica of TG and OG were found cirsoid markedly comparing with SOG( P 〈0.01), the weight of left testicle in TG was intervened between the OG and SOG ( P 〈0.05). The changes of histology were extra marked both in TG and OG, The apoptosis index of TG was lower than that of OG( P 〈 0.05), and higher than that of SOG(P〈 0.05). Conelusions:h shows that α-tocopherol can decrease theapoptosis index in germ cells of varicocle rats, and slow down the proceeding of testicular injured.
