摘要
目的:研究粉防己碱(Tet)对大鼠实验性自身免疫性脑脊髓炎(EAE)的防治作用。方法:采用ic、sc由豚鼠全脊髓匀浆和完全弗氏佐剂制成的抗原乳剂建立EAE模型;Wistar大鼠随机分为正常、模型、Tet组,正常、模型组自免疫当日起均每日ip 2 mL生理盐水,Tet组自免疫当日起每日ip Tet 100 mg·kg^(-1);观察Tet对EAE大鼠发病程度和血清干扰素-γ(IFN-γ)、白细胞介素-12(IL-12)的影响。结果:与模型组比较,Tet组能显著减轻EAE大鼠发病程度(P<0.05),IFN-γ水平降低了41.02%(P<0.05),IL-12水平降低了47.88%(P<0.01)。结论:Tet对大鼠EAE有一定的防治作用,其机制可能与降低血清IFN-γ、IL-12水平有关。
OBJECTIVE: To evaluate the preventive and therapeutic effect of ttrandrine on experimental autoimmune encephalomyelitis (EAE) in rats. METHODS: Wistar rats were injected with antigen emulsion made from spinal cord ho- mogenate together with complete Freund's adjuvant to establish EAE model. Then the model rats were randomized to normal group, model group and Tet group.The rats in normal group and model group were injected peritoneally with 2 mL normal saline daily and those in Tet group with Tet 100 mg·kg^-1 started from the same day of receiving immunization. The severity of the disease, serum levels of IL- 12 and IFN 7 in Tet - treated rats were observed. RESULTS: As compared with model group, Tet- treatment significantly improved the severity of the disease (P〈0.05), down- regulated serum level of IL-12 by 41.02% ( P 〈 0.05) and down - regulated serum level of IFN -γ by 47.88% ( P 〈 0.01) . CONCLUSION : The preventive and therapeutic effect of tetrandrine on experimental autoimmune encephalomyelitis (EAE) in rats is possibly achieved by inhibiting serum levels of IFN -γ
and IL - 12.
