摘要
目的观察盐酸法舒地尔对脑缺血再灌注大鼠神经元损伤的保护作用。方法建立脑缺血再灌注大鼠模型,采用Caspase-9mRNA原位杂交技术,DAB显色,光镜观察细胞浆呈棕黄色为阳性细胞。经图像分析阳性细胞的数量,观察盐酸法舒地尔对脑缺血再灌注损伤大鼠神经元的保护作用。结果盐酸法舒地尔治疗组与模型组比较,Caspase-9mRNA阳性细胞平均密度减小(模型组4.83±0.49,治疗组2.2±0.30),平均灰度值增大(模型组123±7,治疗组191±8),组间比较有明显差异(P<0.05)。结论盐酸法舒地尔对脑缺血再灌注损伤大鼠神经元有一定的保护作用。
Objective To observe the protective effect of fasudil hydrochloride on neuron injury in ischemia reperfusion rats.Methods The ischemia reperfusion injury model was built in rats. The Caspase-9 mRNA hybridization in situ was applied, and DAB coloration, then the neuron was observed by light microscope. The cell pulm presented yellow color was defined positive. The protective effect of fasudu hydrochloride on neurons was observed through quantitative analysis of positive cells. Results Compared with control group, the average density of Caspase-9 mRNA positive cells was decreased in Fasudil group (4.83 ± 0.49 vs 2.2 ± 0.30), and increased in average grave value ( 123 ± 7 vs 191 ± 8). The statistical result was positively ( P 〈 0.05)Conclufion It provided that the fasudil hydrochloride had protective effect on neurons in cerebral ischemia reperfusion rats.
