摘要
对岩藻聚糖的提取工艺进行了优化,并对该多糖的化学成分进行了分析。研究结果发现,岩藻聚糖提取的最佳工艺为:提取温度为90℃,加水量(W/V)为1∶25,提取时间为4h,提取时pH为3.5。在最佳条件下制备得到的岩藻聚糖,经纯化后分析其化学成分,结果表明,蛋白质含量6.9%,重金属元素As和Hg的含量分别为2.26mg/kg、0.119mg/kg,糖醛酸含量为17.2%,用气相色谱分离鉴定单糖成分,该多糖由L(-)-岩藻糖、D-木糖、D-甘露糖、D-葡萄糖、D-半乳糖组成,各单糖含量为40.62%、2.76%、25.88%、22.10%、8.64%(w/w)。
The procedure of extracting fucoidan from kelp has been optimized in this paper, and its chemical components have been analyzed subsequently.It was indicated that the optimal parameters of extracting fucoidan were temperature at 90℃, the ratio of kelp powder weight and water volume 1:25, time 4h, pH at 3.5. With the optimized procedure, the crude fucoidan was made and purified.As for its chemical components, protein was 6.9%, total As and Hg were 2.26mg/kg, 0.11gmg/kg respectively, and the uronic acid was ]7.2%.The monosaccharide composition of fucoidan was analyzed by GC, and this polysaccharide was made up of L(-)- fucose, D-xylose, D-mannose, D-glucose, D-galactose,and their contents were 40.62% ,2.76% ,25.88% ,22.10%, 8.64% ( w/w), respectively.
