摘要 目的:利用近红外光谱(NIR)结合OPUS软件直接测定山药中尿囊素的含量。方法:对山药样品进行近红外光谱检测并采用高效液相色谱(HPLC)法测定其尿囊素的含量,结合OPUS软件建立NIR光谱特征值与HPLC测定的结果之间的校正模型,进而对预测集样品进行分析。结果:校正样品集经内部交叉验证建立校正模型,内部交叉验证决定系数R2=95.47,对预测集样品进行外部验证,预测值与真实值的相关系数R2=95.98。结论:利用NIR技术测定山药样品中尿囊素的含量是可行的,为山药药材的质量控制提供一种快速简便的检测方法。 OBJECTIVE To determine directly the allantoin in Rhizoma Dioscoreae by combining Near-infrared Spectroscopy (NIR) and OPUS software. METHODS Samples with different concentrations were analyzed by NIR spectra and HPLC. The OPUS software was used to establish the calibration model between the information of NIR and HPLC. Furthermore, tests of the best fitted model with prediction samples were made. RESULTS The internal cross certification decision coefficient of correcting samples through internal cros-certification was 95.47 to establish calibration model. The correlation coefficient of the prediction samples for external validation between the predictive value and the real value was 94. 99. CONCLUSION NIR technique can be used in de termination of allantoin in Rhizoma Dioscoreae and can providea rapid, simple on-line detection method for Rhizoma Dioscoreae.
