摘要
用培养基发芽法研究沙芥花粉萌发特性;用人工异株授粉法和联苯胺—过氧化氢法测定柱头可授期。结果表明:沙芥花粉萌发的最佳培养基是20%蔗糖+0.001%硼酸+0.5%琼脂;花粉能够萌发的温度范围为15~38℃,最适萌发温度为25℃;从花蕾充分膨大期至开花后16h花粉活力较强,开花后48h花粉基本无活力;花粉储藏的适宜条件为低温(4℃),储藏期为7d。柱头可授期为开花前48h至开花后96h,开花后24~72h可授性强。
Pollen germination characteristic of Pugionium cornutum ( L. ) Gaertner was evaluated by germination method on culture medium, its stigma receptivity was estimated by the hand cross-pollination and benzidine - H2O2 method. The results showed the optimal pollen germination of P. cornutum was on the culture medium added 20% sucrose, 0. 001% boric acid and 0. 5% agar. The effective temperature of pollen germination was from 15 ℃ to 38 ℃ and the optimal temperature was 25 ℃. The pollen germination rate was very higher from the biggest bud stage to 16 h after blooming, and the pollen viability was disappeared 48 h after blooming. The suitable condition of storing pollen was low temperature (4 ℃ ), and storing period was 7 d. The stigma of P. cornutum had receptivity from 48 h before blooming to 96 h after blooming, the strong receptivity in 24 -72 h after blooming.
