摘要
采用PCR-SSCP技术,对中国荷斯坦乳牛DRB3基因的多态性进行了分析,并用线性数学模型分析了各种基因型与乳牛乳腺炎的相关性。结果显示,DRB3基因有AA、BB和AB 3种基因型,其基因型频率分别为0.46,0.33和0.21;DRB3基因多态的产生是第88位产生A→T突变、第118位产生T→A突变以及第165位产生A→G的替换造成的;基因型频率分布的2χ值为19.56(P<0.01),DRB3基因的突变处于Hardy-Weinberg非平衡状态。相关性分析表明,不同基因型乳牛之间泌乳性状的差异不显著。
Polymorphism in DRB3 gene in Chinese Holstein dairy cow was analyzed by PCR-SSCP. The association of genotype with mastitis was analyzed by the linear mathematical model method. The correlation of DRB3 genotype with lactation traits was analyzed. There were three genotypes of AA, BB and AB in the DRB3 gene site of Chinese Holstein dairy cow and their genotype frequencies were 0.46,0.33 and 0.21 respectively. The polymorphisms of DRB3 gene were caused by A→T mutation at the 88 bp posi- tion,T→A mutation at the 118 bp position and A→G transversion at the 165 bp position. The zetest indicated that the Xe value among genotype frequencies of DRB3 gene site was 19.56(P〈0.01),indicating that mutations of DRB3 gene in the Chinese Holstein dairy cow were out of Hardy-Weinberg Equilibrium. The correlation analysis showed that the effect of DRB3 genotypes on lactation traits was not significant.
