摘要
目的了解实验性视网膜挫伤后神经感觉层细胞凋亡情况,并进一步探讨凋亡的发生机制,为临床治疗提供理论基础。方法40只健康成年无眼疾青紫兰兔,随机数字表法分为挫伤后1h、3h、1d、3d、7d、14d、1个月及正常对照组8组,每组5只,选取右眼为致伤眼,以改良Allen重击法制备兔单眼挫伤性视网膜病变模型;分别于挫伤后1h、3h、1d、3d、7d、14d、1个月获取兔眼标本,以电镜及TUNEL法观察视网膜神经感觉层细胞的凋亡情况。结果视网膜挫伤后存在着神经感觉层细胞凋亡现象,正常对照组、14d和1个月组几乎不见凋亡细胞,挫伤后3h、1d、3d在视网膜各细胞层均出现较多的、具有凋亡形态学与生化改变特征的凋亡细胞,其中在3 d组视网膜感觉层凋亡细胞数量达到高峰。视网膜神经感觉层细胞数目在3h、1d、3d组及7d组之间两两比较均有显著统计学意义(P<0.01);3h、1d、3d组及7d组与正常对照组、1h、14d、1个月组之间两两比较均有显著统计学意义(P<0.01);正常对照组、1h、14d、1个月组之间两两比较无显著性意义(P>0.05)。结论视网膜挫伤后,神经感觉层细胞凋亡的发生是视网膜功能恢复不良的重要原因。
Objective To observe the cell apoptosis of sensory layer after contusive injury in rabbit retina,and further to study the apoptosis mechanisms providing the rationale for clinical treatment. Methods Forty healthy rabbits without oculopathy were divided into 8 groups, 1 hour,3 hours, 1 day,3 days,7 days,14 days, 1 month after contusion and normal control, 5 rabbits in each group. Right eye of each rabbit was treated with Allen' s reformative hitting method as contusive retinopathy model. The eyeballs were enucleated at different times after contusive injury in rabbit retina. The morphologic changes of the retina were observed by electron microscope. Terminal deoxynueleotidyl trans- ferase-mediated dUTP nick end labeling technique was used to assess apoptosis. 6esults There was cell apoptosis in sensory layer after contusion of retina,but almost not observed in retinas from control, 14 days and 1 month eyes. They were abundant at 3 hours, 1 day and 3 days after contusion of retina,peak- ed at 3 days,and dropped precipitously at 7 days after contusion. Every multiple comparison among the groups of 3 hours, 1 day, 3 days and 7 days after retinal contusion showed significant difference in comparing of apoptotic cells ( P 〈 0. 01 ) ;An obvious change of cell number was found comparing each of 3 hours, one day,3 days and 7 days with each of normal control, one hour, 14 days and one month( P 〈0.01 ). But there was no significant difference of multiple comparison among control group, 1 hour, 14 days, and 1 month group after retina contusion( P 〉 0.05 ). Conclusion Apoptosis occurs in sensory layer after contu- sion of retina, which may lead to incomplete recovery of retinal function. [ Rec Adv Ophthalmol 2008 ;28 (8) :590-594 ]
