摘要
目的探讨四逆汤对氧自由基所致心肌细胞L型钙电流异常的作用。方法用急性酶解法获得单个大鼠心室肌细胞,用标准的全细胞膜片钳技术记录钙通道电流,首先观察5mmol/LH2O2对心肌细胞ICa,L的改变,然后观察预先用四逆汤含药血清孵育30min后5mmol/LH2O2对心肌细胞ICa,L的影响。结果在H2O2作用下,大鼠心室肌细胞ICa,L电流密度增加,电流-电压曲线显著下移,稳态失活曲线明显左移。四逆汤含药血清对离子电流无直接影响,但可以减轻H2O2所引起的稳态失活曲线的异常改变。结论四逆汤可减轻H2O2对心肌细胞钙通道的损伤作用。
Objective To determine the effect and mecha-nism of Sini Decoction on H2O2 Induced L-type calcium current (ICa,L) changes in single rat vemtricular myocyte. Methods Whole-cellpacth-clamp recording technique was used to record ICa,L in single cells isolated rat ventricular myocytes. Results The nifedipine-sensitive L-type calcium inward current was recorded in the rat ventricular myocytes 5 mmol/L H2O2 increased the calcium channel current in a voltage-dependent manner in ventricular myocytes of rats. The current-voltage curve was shifted downwards and the inactivation curve was shifted to more negative. The serum of Sini Decoction had no effect on current-voltage curve. However, it relieved the H2O2 induced inactivation curve changes. Conclusion Sini Decoction can relieve H2O2 induced L-type calcium channel current injury.
