α-生育酚清除神经元细胞内外氧自由基作用观察 被引量:1

摘要 目的观察α-生育酚对神经元细胞内外氧自由基的清除作用。方法培养SD胎鼠皮层神经元细胞,将神经元细胞分为正常对照组、单纯氧自由基损伤组和α-生育酚保护组。用Fenton反应造成神经元细胞损伤,用激光共聚焦显微镜观察各组神经元DCFH-DA染色情况,确定神经元细胞内氧自由基浓度,并用ESR法检测各组神经元细胞外液中的氧自由基浓度。结果α-生育酚保护组神经元细胞内氧自由基浓度在不同时间点均低于单纯氧自由基损伤组(P均<0.05),神经元细胞外氧自由基浓度在不同时间点亦均低于单纯氧自由基损伤组(P均<0.05)。结论α-生育酚可以有效清除神经元细胞内外的氧自由基。 [ Objective ] To evaluate the effect of α-tocopherol eliminating intraeellular and extraeellular oxidative free radicals of neurons. [ Methods ] Culturing the neurons of the cortex of SD embryo mice. Divided the neurons into three groups: normal group, oxidative damage group and α-tocopherol protection group. Induced the oxidative damage to oxidative damage group and α-tocopherol protection group by Fenton reaction. Dyed the neurons with DCFH-DA, and observed the concentration of intracellular oxidative free radicals of neurons with confocal laser scanning microscope. The concentrations of oxidative free radicals in extracellular solution of different groups was detected by ESR. [ Results ] α-Tocopherol reduced the concentration of intracellular( P 〈 0.03 ) and extracellular( P 〈 0.03 ) oxidative free radicals of neurons. [ Conclusion ] α-tocopherol can eliminate the intracellular and extraeellular oxidative free radicals of neurons effectively.
