喂饲前体提高悬浮培养水母雪莲细胞的黄酮产量 被引量:5

Increased flavonoids production in suspension culture of Saussurea medusa cells by the addition of precursors

摘要 为提高水母雪莲细胞悬浮培养的生物量及次级代谢产物黄酮的含量,在培养基中喂饲肉桂酸、乙酸钠和苯丙氨酸等三种黄酮生物合成前体,考察了它们对水母雪莲细胞TUIP-8悬浮培养的影响。在(17±1)℃环境中悬浮培养TUIP-8水母雪莲细胞,喂饲这三种前体均能显著促进雪莲黄酮的生物合成,并且对细胞的生长抑制效应较小。在接种时喂饲前体,低浓度添加肉桂酸(7.14mg/L)、乙酸钠(3.57mg/L)和苯丙氨酸(3.57mg/L)更有利于提高黄酮产量。培养后期喂饲前体的效果要好于接种时喂饲,其中17.85mg/L的乙酸钠对黄酮合成的促进作用最强,可使黄酮含量从7.8%提高至12.4%,黄酮产量可提高63%。在(25±1)℃环境中悬浮培养TUIP-8水母雪莲细胞,喂饲前体促进雪莲黄酮生物合成的效果不明显,与对照相比黄酮产量只提高5%-10%。反应器培养的雪莲细胞总黄酮含量可下降至8.5%,但在培养后期使用含有28mg/L乙酸钠、60mg/L苯丙氨酸的改进MS培养基,雪莲细胞的黄酮含量可恢复至11%,细胞密度可达到21g(干重)/L。在悬浮培养水母雪莲细胞过程中添加乙酸钠和苯丙氨酸等前体物质可以促进雪莲黄酮的合成。 The effects of feeding three flavonoid precursors (cinnamic acid, NaAc and L-phenylalanine) on suspension culture of Saussurea medusa cells were investigated to increase the biomass of cultivated Saussurea medusa cells and the production of flavonoids. The results showed that the addition of the three precursors could enhance flavonoid biosynthesis signif- icantly when the TUIP-8 cells, a stable cell line derived from S. medusa callus, were cultivated at ( 17 ± 1 ) ℃, and had little inhibitory effect on cell growth. Feeding of lower concentration of cinnamic acid (7.14 mg/L), NaAc (3.57 nag/ L) and L-phenylalanine (3.57 mg/L) was more beneficial to increasement of production of flavonoids when the precursors were added at inoculation. The precursors' feeding in later duration of the experiment was more effective in enhancement of flavonoid formation than at inoculation. Among the three precursors, NaAc had the most significant promoting effect on flavonoid biosynthesis at the concentration of 17.38mg/L, and the flavonoid content could increase from 7.8% to 12.4% and flavonoid production could raise 63% as compared to the control. The feeding of precursors had a little enhancement effect in flavonoids production (5% - 10%) when TUIP-8 cells grew at 25℃. Without feeding precursors, the flavonoids in S. medusa cells suspended culture in a 8L stirred tank dropped dramatically to 8.5% ; whereas, the flavonoid content could get back to the 11% when the cells grew in the reactor with the modified MS medium containing 28mg/L NaAc and 60mg/L L-phenylalanine during the later culture period, and the cell density could reach 21g (dry weight)/L. In conclusion, the addition of precursors such as NaAc and L-phenylalanine can effectively promote flavonoid biosynthesis in suspended culture of Saussurea medusa cells.
