Screening of serum biomarkers in acute leukemia by protein chip

摘要 目的利用蛋白质芯片从初诊急性白血病患者血清中筛选标记蛋白质。方法采用表面增强激光解吸电离飞行时间质谱技术(SELDI-TOF-MS surfaced enhanced laser desorption/ionization time of flight mass spectroscopy)和弱阳离子交换(WCX2)蛋白质芯片对80例初诊急性白血病患者(44例儿童急性淋巴细胞白血病、36例成人急性粒细胞白血病)血清和78例正常人(35例为儿童)血清的蛋白质图谱进行检测,使用PBSII-C型蛋白质芯片阅读机读取数据。结果急性淋巴细胞白血病初诊患儿与正常儿童血清蛋白质谱相比有显著差异蛋白质7个,用标记分子建立儿童急性淋巴细胞白血病诊断的分类树模型,检测灵敏度为88.6%(39/44)、特异性为82.9%(29/35);成人急性粒细胞白血病患者与正常成人血清蛋白质谱相比有显著差异蛋白质8个,用其中的7个标记分子建立成人急性粒细胞白血病诊断的分类树模型,检测灵敏度为86.1%(31/36)、特异性为95.3%(41/43)。结论蛋白质芯片技术对于发现和筛选急性白血病血清中的标记蛋白质是一种有效、快速的工具。 Objective To screen serum biomarkers in patients with acute leukemia. Methods Proteomie spectra were generated by mass spectrometry in 158, including 44 acute lymphocyte leukemia patients and 36 acute myeloid leukemia, 78 healthy control. 158 spectra obtained were used to train and to develop a decision tree classification algorithm. Results A total of 7 distinguished proteomic peaks were detected in acute lymphocyte leukemia and were used to build a proteomic pattern. The results yielded a sensitivity of 88.6% (39/44) ,specificity of 82.9% (29/35) ;A total of 8 distinguished proteomic peaks were detected in acute myeloid leukemia,7 peaks were used to build a proteomic pattern. The results yielded a sensitivity of 86. 1% (31/36), specificity of 95.3% (41/43). Conclusion SELDI-TOF-MS offers a unique platform for the proteomic detection of acute leukemia. It also offers a noninvasive method to further study the proteomic changs in the development and progression of leukemia.
