摘要
目的:探讨α-干扰素(IFN-α)和γ-干扰素IFN-γ对淋巴管内皮细胞增殖和游走的影响及作用机制。方法:内皮细胞取自猪的胸导管。应用Hoechst和Caspase染色法检测IFN-α和IFN-γ诱导的细胞凋亡;采用划线法和MTT法观察IFN-α和IFN-γ对淋巴管内皮细胞增殖和游走的影响;活兔后肢切断淋巴管并分别注射IFN-α和生理盐水,观察淋巴管的再生情况。结果:经IFN-α和IFN-γ处理后,淋巴管内皮细胞其核周围有凋亡小体;Caspase染色阳性表达;IFN-α和IFN-γ对淋巴管内皮细胞的增殖和游走有明显的抑制作用;手术切断兔后肢淋巴管后16d,IFN-α侧仍有大量染料渗漏。结论:IFN-α和IFN-γ具有促进细胞凋亡的作用。IFN-α和IFN-γ对淋巴管内皮细胞的增殖和游走有明显的抑制作用。
Objective: To study the effect and mechanism of IFN-α and IFN-γ on proliferation and migration of lymphatic endothelial cells(LEC). Methods: LECs were taken from the pig thoracic duct. Hoechst and Caspase staining was applied to examine apoptosis of LECs. Method of scraping line and MTT were used to detect IFN-α and IFN-γ on proliferation and migration of LECs. Lymphatics of two posterior limbs in rabbits were cut and IFN-α and 0.9% NaCl solution were injected around the lymphatic vessels of each limb respectively to observe healing of the lymphatics. Results: After the treatment with IFN-α and IFN-γ, the apoptotic body was found surrounding the nucleus of LECs. Caspase staining proved the positive cells of apoptosis. IFN-α and IFN-γ could evidently inhibit proliferation and migration of LECs. IFN-α experiment in vivo : 16 days after the operation, the lymphatic vessels which had been injected with IFN- α solution had a serious leak. Conclusion: IFN-α and IFN-γ can promote apoptosis of LECs and evidently inhibit the proliferation and migration of LECs.
