甘肃猫儿眼多糖对SMMC-7721细胞毒性机制研究 被引量:5

摘要 给予人SMM C-7721细胞甘肃猫儿眼多糖(浓度为10.0、1.0和0.1μg/mL)48 h,用四唑盐(MTT)法、台盼蓝拒染法和流式细胞仪进行检测.结果表明,甘肃猫儿眼多糖对人SMM C-7721细胞增殖的抑制率分别为72%、47%、31%;活细胞数为12.7%、28.6%、68.9%;细胞调亡率为26.4%、18.2%、16.3%.结果显示,甘肃猫儿眼多糖具有明显的细胞毒性和抑制细胞增殖、诱导细胞凋亡的功能,其机制与破坏细胞膜结构,损伤线粒体和DNA,抑制细胞分裂有关. The human SMMC-7721 strain cells were given with maoeryan polysaccharide in vitro at the concentrations of 10.0,1.0 and 0. 1 μg/mL for 48 h, respectively. The cytotoxic effect, the inhibitory rate for proliferation of SMMC-7721 strain cells and the apoptosis rate were determined by using flow cytometry, methods of MTT [3-(4,5-dimethyl thiazol-2yl)-2,5-diphenyhetrazolium bromide] and trypanblue. The results showed that the maoeryan polysaccharide had significant cytotoxic effects on human SMMC-7721 strain cells. The inhibitory rates wore 72%,47%,31%, and the apoptosis rate 26.4%, 18. 2%, 16.3%, respectively, as compared with those of the control group. The maoeryan polysaccharide has significant cytotoxic effects on human SMMC-7721strain cells and their mechanism -nay be related to the inhibitation of cell division, and to the destruction of cell membrane and mitochondria.
