Expression of hMLH_1 gene in gastric cancer and its significance

摘要 目的:探讨错配修复基因hMLH1在胃癌组织中的表达及其意义。方法:采用免疫组织化学和逆转录-聚合酶链反应(RT-PCR)检测68例胃癌组织、33例癌旁组织和35例正常胃组织中hMLH1蛋白和mRNA的表达情况,同时采用原位末端标记法测定癌组织的细胞凋亡指数(AI)。结果:胃癌组织、癌旁组织和正常胃组织hMLH1mRNA表达率分别为63.3%(43/68)、100%(33/33)和100%(35/35),蛋白表达率分别为61.8%(42/68)、93.9%(31/33)和100%(35/35),hMLH1表达与胃癌组织的临床分期呈正相关(r=0.513,P<0.05)。hMLH1表达阳性胃癌组织AI为(3.652±0.241)%,与hMLH1表达阴性胃癌组织AI[(1.887±0.212)%]比较差异有显著性(P<0.01)。结论:hMLH1在胃癌组织中低表达,提示错配修复基因hMLH1功能缺陷在胃癌的发生和发展过程中起重要作用。 Objective To investigate the expression of hMLH1 in gastric cancer and its significance, Methods The expressions of hMLH1 protein and mRNA were detected by immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR) in 68 cases of gastric cancer tissues, 33 cases of para-cancer tissues and 35 cases of normal gastric tissues. Apoptosis was assayed by terminal deoxytransferase mediated dUTP nicked labeling. Results The positive expression rates of hMLH1 mRNA were 63.3% (43/68), 100% (33/33) and 100% (35/35), and those of protein were 61.8% (42/68), 93.9% (31/33), 100% (35/35), respectively, in gastric cancer tissues, para-cancer tissues and normal gastric tissues. There was a correlation between the expression of hMLH1 and clinical grade of gastric cancer (r=0. 531, P〈0.05) . The apoptotic index in the gastric cancer tissue with positive expression of hMLH1(3. 652 ± 0. 241)%] was higher than that in the gastric cancer tissue with negative expression of hMLH1 [ (1. 887±0. 212)%] (P〈0. 01). Conclusion The low expression of hMLH1 in gastric cancer tissue indicats that hMLH1 expression may play an important role in the pathway of carcinogenesis of gastric cancer.
