摘要
建立了同时测定枸杞中类胡萝卜素及其酯类化合物含量的高效液相色谱分析方法,采用ODS-C18色谱柱(250mm×4.6mm;5μm),流动相为甲醇-乙腈-二氯甲烷-正己烷(15:40:20:20,V/V)等度洗脱,二极管阵列检测器,检测波长为450nm,流速1.0mL/min,柱温25℃。外标法定量,玉米黄素、β-胡萝卜素、玉米黄素双棕榈酸酯3组分线性关系良好,相关系数0.9991~0.9998;平均回收率为98.8%~100.5%;RSD为0.8%~1.8%;检出限为0.02~0.20mg/L。方法简便、快速、准确,可以用于枸杞类胡萝卜素定量测定。
A method was developed for the determination of carotenoids and carotenoid esters in Fructus Lycii by reversed-phase high performance liquid chromatography. Separation and quantification were achieved by an ODS-C18 column (250 mm×4.6mm; 5 μm), methanol-acetonitrile-dichloromethane-hexane (15:40:20:20, V/V) as mobile phase at a flowrate of 1 mL/min, temperature 25 ℃ and detected with photodiode array detector at 450 nm. The quantified analysis of external standard calibration curves was used. There were good linear correlations between the concentrations of zeaxanthin, E-carotene, zeaxanthin dipalmitate and their chromatographyic peak areas. The correlation coefficients were 0.9991 - 0.9998. The average recoveries were 98.8% - 100.5% with RSD of 0.8% - 1.8%. The detection limits were 0.02 - 0.20 mg/L. This method is simple, fast, accurate and convenient, it is one of the means for the determination carotenoids in Fructus lycii quality mensuration.
