摘要
目的 建立胶束电动毛细管电泳色谱法(MECC)测定半夏及其同科其它品种药材中苯甲酸的含量的方法。方法 用MECC法对样品进行分析。毛细管柱内径75μm,长62.5cm;运行电压25kV,检测波长200nm,分离温度35℃;缓冲液为30mmol·L^-1,SDS-30mmol·L^-1硼酸-15%甲醇(pH值9.5);压力进样,进样时间3s。结果 苯甲酸在0.044-2.2mg/ml浓度范围内具有良好的线形关系(r=0.9994),平均回收率为96.7%(RSD=2.4%,n=6),对半夏及其同科属其它品种共14个样品中的苯甲酸含量进行了测定。结论 该方法快速、简便、并且较为准确,可供评价半夏等品种质量时参考。
Objective The method of micellar electrokinetic capillary chromatography (MECC) was established to determine the contents of benzoic acid in Rhizoma Pinelliae and several Rhizomes from other Araceae species ; Methods The samples from rhizomes of P. ternana and other species of Araceae were analyzed by MECC; The Capillary column was 62. 5 cm long and 75 μm in internal diameter; The operation voltage 25 kV ; test wavelength 200 nm; column temperature 35 ℃ ; the mobile buffer was 30 mmol· L^-1 SDS-30nunol · L^-1 boric acid -15% methyl alcohol (pH 9. 5) ; The sampling was conducted by pressure, the sampling time was 3 seconds. Results The linear range of determination for benzoic acid was 0. 044 - 2.2 mg,/ml ( r = 0. 999 4). The average recovery rate was 96, 7% (cv = 2.4%, n = 6). The contents of benzoic acid in the 14 samples from P. ternana and other species of Araceae were determined. Conclusion The method is fast, simple, convenient, comparatively accurate, and can be used to refer when appraising quality of Rhizoma Pinelliae and several species.
