恶性血液病RAS基因突变激活与Ras蛋白法尼基化抑制剂的研究进展 被引量:2

摘要 RAS癌基因是最早被确定的由点突变方式激活而致细胞癌变的原位癌基因,近年来研究发现RAS基因家族可通过受体介导的信号传递参与多种血细胞的增生、分化过程,导致多种恶性血液病的发生。如何抑制Ras信号通路调控血细胞的增殖与分化已成为研究的热点之一。法尼酰基转移酶抑制剂通过抑制法尼基蛋白转移酶阻止Ras蛋白的法尼基化、裂解和羧甲基化修饰,使其失去生物活性,从而抑制了Ras信号传导通路,已成为新的很有希望的药物作用靶点。
