摘要
综述了RAPD在昆虫分类学、系统发育分析、种群遗传学、连锁图谱的构建、特殊基因的检测和定位、父系分析、精子竞争以及杂交带的确定等研究中的应用。目前,在昆虫学研究中,因RAPD的致命缺点使人们对其的兴趣有所降低,而把更多的注意力转向一些更可靠的如微卫星等共显性标记,但各种分子标记都有一定的缺陷,RAPD仍然会是一种重要的分子标记。在我国,RAPD被大量地应用于昆虫学的研究中,并取得了丰硕成果,但在样本量、扩增条件优化、数据分析、研究问题的深度及应用范围等方面还有待进一步发展。
Random amplified polymorphic DNA(RAPD) was developed in the 1990' s, was heap, easy to develop,needed no prior information of template DNA,so it had been widely used for many types of genetic analyses in entomological studies. A review on the applications of RAPD is given for the following entomological investigations: identification of various taxa,phylogenetic analyses,description of population genetics,construction of linkage and physical maps, location of spec:ial genes, parentage analyses, sperm contention and determination of hybrid zones. More attention has been focused on some reliable codominant molecular markers such as microsatellite, however, every molecular marker has its limitations. In China, RAPD has been widely used in entomological studies and has given many useful results, more improvements should be performed toward sampling size, the optimization of reaction conditions, data analysis and the selection of profound study objects when using RAPD in entomology.
