摘要
目的观察肝癌小鼠模型射频治疗后外周血肿瘤坏死因子-α(TNF-α)和白细胞介素-2(IL-2)的变化及癌组织 CD54的表达情况,探讨肝癌射频治疗对机体免疫功能的影响。方法 Balb's小鼠60只,随机分为2组,对照组(C 组)和射频治疗组(R 组),每组30只。成功制作小鼠肝细胞癌模型后,R 组给予射频治疗,并于其后0h、2h、2周、3周、4周处死并眼球取血法取血,采用双抗体夹心 ELISA 法测定 TNF-α和 IL-2光密度值(OD),同时切除肿瘤进行免疫组化 CD54表达分析。结果R组肿瘤明显缩小,其 TNF-α和 IL-2浓度呈递增趋势,与对照组比较差异有显著性(P<0.05);经射频处理后的 R 组肝癌细胞 CD54呈过度表达现象,并与各时间段 TNF-α、IL-2浓度呈明显正相关(r=0.520,P<0.01;r=0.501.P<0.01)。结论肝癌射频治疗诱使 TNF-α、IL-2浓度增高,并上调细胞间粘附分子 CD54的表达,从而诱导肿瘤免疫应答而发挥抗肿瘤作用。
Objective To investigate the effects of radiofrequency on immune function of mice with hepatocellular carcinoma (HCC). Methods Sixty Balb's mice were randomized into the control group (groups A, n=30) and radiofrequency-treated group (group B, n=30). After the model of HCC were established in all the 60 mice, radiofrequency treatment was performed in group 13. Blood samples were harvested by eyeball blood collection at Oh, 2h, 2w, 3w and 4w after the treatment. The changes in TNF-α and IL-2 were determined by ELISA and that in CD54 by s-p immunohistochemical staining. Results The levels of TNF-α and 11-2 was significantly higher in group B than in group A (P〈0. 05). In group B, the expression of CD54 was increased and it was correlated with the changes of TNF-α and 11-2 levels (P〈0. 01). Condusions Radiofrequency treatment can increase the blood levels of TNF-α and 11-2 and up-regulate the expression of intracellular adhesion molecule to induce immune response of the tumor to exert anti-tumor effect.
