应激性肾上腺素与动脉粥样硬化相关机制研究

摘要 目的观察心理应激时肾上腺素对THP-1源性巨噬细胞脂质转运相关基因三磷酸腺苷综合盒转运体A1(ABCA1)、小凹蛋白(Caveolin-1)与细胞因子转化生长因子(TGF-β1)mRNA表达的影响,探讨应激性肾上腺素在动脉粥样硬化中的作用。方法不同浓度肾上腺素(1pmol/L-1μmol/L)处理THP-1源性巨噬细胞24h,运用逆转录多聚酶链反应检测其ABCA1、Caveolin-1与TGF-β1mRNA表达。结果1pmol/L-10nmol/L的肾上腺素对受试基因mRNA的表达与相应对照组比较其差异无显著性(P>0.05)。而100nmol/L及1μmol/L的肾上腺素下调TGF-β1和ABCA1mRNA水平,与各自对照组比较其差异有显著性(P<0.05);但100nmol/L及1μmol/L的肾上腺素对Cave-olin-1mRNA的表达无明显影响,与对照组比较其差异无显著性(P>0.05)。结论肾上腺素下调THP-1源性巨噬细胞TGF-β1与ABCA1mRNA水平,不影响Caveolin-1mRNA表达。 Objective To investigate the effect of adrenaline on the expression of ABCA1 mRNA,Caveolin- 1 mRNA and TGF-β1 mRNA in THP- 1 macrophages. Methods THP - 1-derived macrophages were incubated with various concentra tions ( 1 μmol/L to 1 μmol/L) of adrenaline for 24 hours. Then ABCA1, Caveolin-1 and TGF -β1 mRNA expression were determined by RT-PCR. Results The different concentrations( 1 μmol/L to 10 μmol/L) of adrenaline had no significant effect on these genes expression (P 〉0.05), but the adrenaline of higher concentrations ( 100 μmol/L, 1 μmol/L) significantly decreased the expression of ABCA1 mRNA and TGF- β1 mRNA except Caveolin-1 mRNA ( P 〈 0.05 ) compared with the control (without adrenaline). Conclusions Adrenaline can down-regulate the expression of ABCA1 mRNA and TGF-β1 mRNA,but not Caveolin-1 mRNA, in THP-1 macrophages.
